In vitro transcription of mouse mammary tumor virus RNA into DNA.

چکیده
The in vitro transcription of the B-type mouse mammary tumor virus RNA into DNA is similar to that obtained with certain C-type viruses: synthesis can be initiated on an RNA primer molecule approximately the size of a tRNA, and a unique linkage (rA-dC) exists between this primer and the DNA product.

Transcription initiation of transfected mouse mammary tumor virus LTR DNA is regulated by glucocorticoid hormones.
A chimeric gene, recombined in vitro, containing a long terminal repeat (LTR) sequence from the proviral DNA of mouse mammary tumor virus and the thymidine kinase (tk) gene of Herpes Simplex Virus was introduced into L tk- cells. No transcription of LTR RNA was observed in transfected cells when glucocorticoid hormones were absent from the growth medium. Accumulation of LTR initiated RNA was me...

A cat kidney cell line, CRFK-F2, was successfully inoculated in suspension and in monolayer culture with a purified mouse mammary tumor virus derived from RIII milk. The virus produced by the infected cells was identified by immunogluorescence, electron microscopy, and RNA-directed DNA polymerase assays; it was a B-type virion that did not cross-react with mouse or feline leukemia-sarcoma virus...

The mouse mammary tumor virus long terminal repeat (MMTV-LTR) participates in the control of gene expression by providing a series of important DNA binding sites at which trans-acting factors interact. Among these factors are the steroid receptor, nuclear factor I (NFI) and the TATA box factor (TFIID). The binding of these proteins facilitates the assembly of a transcriptionally competent compl...

Expression of mouse mammary tumor virus in T lymphocytes appears to be required for accession of horizontally transmitted virus to the mammary gland. Further, deletions in the long terminal repeat which relax constraints on viral transcription promote T cell lymphoma. We have identified a polypurine transcriptional repressor element (NRE1) that is deleted from viruses that induce T cell lymphom...
متن کاملChanges in retrovirus expression in mouse mammary tumor cells in culture.
Mouse mammary tumor cells (Mm5Mt/c1) produced mouse mammary tumor virus (MMTV) (type B retrovirus) at early passages in culture. They apparently produced predominently type C retrovirus(es) at late passages in culture. The decline in MMTV production during passage was not accompanied by an apparent decrease in the synthesis or accumulation of intracellular MMTV-specific RNA. As judged by the co...
